The Carlyle Group to acquire rapidly growing life sciences tools company Unchained Labs

April 26, 2021 – Pleasanton, CA – Global investment firm The Carlyle Group (NASDAQ:CG) today announced that it has agreed to acquire the rapidly growing life sciences tools company Unchained Labs for $435 million, in partnership with its management team, from Novo Holdings A/S, Canaan Partners and TPG Biotech. The transaction is subject to regulatory approvals and is expected to close in Q2 2021.

Founded in 2015 and headquartered in Pleasanton, CA, Unchained Labs is a life science tools company focused on solving problems for biologics and gene therapy researchers. Unchained systems are used by biopharma researchers around the world to analyze and characterize large, complex molecules and to make their research jobs easier. Unchained has 170 employees and expects to generate over $75 million of revenue in 2021.

Unchained Labs makes another big splash with Big Tuna in gene therapy and vaccines!

April 1, 2021 – Pleasanton, CA – Unchained Labs, the life sciences company that’s all about getting researchers the right tool for the job, launched its new gene therapy and vaccine applications on the Big Tuna platform today. Big Tuna is already the ultimate buffer exchange platform for biologics, and now it can take on buffer exchange, concentration and clean-up of adeno-associated viruses (AAVs) and lipid nanoparticles (LNPs).

AAVs are produced at low concentrations and need to be concentrated up for characterization. LNPs are made in the presence of ethanol which needs to be cleared out quickly to keep them stable. Both are tough to handle due to their large size and how fast their buffers flow through filtration membranes. Big Tuna’s new high molecular weight cutoff plates, concentration application and tunable pressure settings make it perfectly dialed in for these molecules.

Unchained Labs launches the ultimate gene therapy tool; tackle AAV titer, empty/full ratio and more with Stunner!

October 6, 2020 – Pleasanton, CA – Unchained Labs, the life sciences company that’s all about getting researchers the right tool for the job, launched its new gene therapy quantification application on Stunner today. Stunner is the first platform that gives researchers a full read-out on how many AAV capsids they have, how many are packed with DNA and if there is aggregation – in less than a minute using only 2 µL of sample.

Adeno-associated viruses (AAVs) used in gene therapies are hard to manufacture and tough to characterize.  Researchers need to be able to quickly and accurately measure their capsid titer, and how many of them are full of DNA they are supposed to deliver to cells. Other methods that gather this info require labels, dyes, reagents, standard curves or use a lot of sample. Pulling it all together is a big lift and takes way too long with existing technologies.

Stunner makes AAV titer a snap. Researchers just pipette 2 µL of their sample, up to 96 samples per run, and then Stunner takes it from there. UV/Vis measures the total protein and DNA in the sample. Dynamic light scattering identifies AAV capsids based on their size and checks for aggregation. Stunner then combines all this information to provide an accurate titer and empty/full ratio.

Uncle is already the gold standard for measuring protein stability and, with this new application, jumps in to support researchers in pursuit of novel gene therapies and better vaccines.

Many gene therapies, immunotherapies and vaccines being developed use adeno-associated viruses or AAVs. These viral vectors are great at sneaking past the body’s immune system to deliver DNA that commands cells to make proteins that end diseases or antibodies that seek out and destroy viruses. A stable AAV stays intact until it finds its target and won’t aggregate – or worse, leak potentially toxic DNA into the body.

Uncle’s newest application helps researchers understand their AAV stability. It uses a thermal ramp with a DNA-binding fluorescent dye to help them get a read on when DNA starts to leak from an AAV, even before the capsid starts to fall apart. Uncle’s DLS and SLS combo can measure the particle titer to see how many capsids they have. They’ll also know when aggregation gets out of hand.

This release is chock-full of new data analysis tools and expands the biologics application space for Uncle, the undisputed all-in-one stability screening platform.

When developing a protein or formulation, aggregation is always a no-go. Right from the start, Uncle’s been giving researchers insight into the connection between protein unfolding and aggregation with its combo of fluorescence and static light scattering. Uncle 4.0 software helps them dig even deeper with a beefed-up dynamic light scattering application that grabs more data points during thermal ramps. The higher resolution that comes with it gives scientists a much closer look at aggregation events.

Uncle’s always delivered the complete protein stability story using full-spectrum fluorescence. The Uncle 4.0 update lets researchers choose exactly what part of the spectrum they want to analyze, so they can focus in on even subtle changes happening to their protein. Being able to dial up a specific wavelength band of interest gives them the option to work with many fluorescent dyes, making it possible to characterize membrane proteins and other tricky molecules.

The agreement allows the company to retire its existing $18.5 million debt facility and complements the $86 million raised in equity over the last four years from Novo Ventures, Canaan Partners, TPG Biotech and Tri-Valley Ventures.

Unchained Labs cuts the ribbon on new contract testing lab in North America!

January 15, 2019 – Pleasanton, CA – Unchained Labs, the life sciences company that’s all about getting biologics researchers the right tool for the job, opened the doors to its new contract testing lab today. The lab is located at the company’s headquarters in Pleasanton, California and provides identification of unwanted particles in drug products and quality assessment of the silicone layers on drug delivery devices.

Two Unchained Labs instruments are heavily used for contract testing. For visible particles, Hound determines the count, size, shape and identity with either Raman or LIBS spectroscopy. If clients have particles in the sub-visible range, the lab has the tools to hunt down those too. Bouncer is used to measure the silicone thickness and distribution on syringes, cartridges and vials.

All sample preparation and testing are handled by trained experts who deliver detailed reports that document the test plan, measurement parameters and results. Turnaround time for results can be as quick as 3 days.

The buffer exchange process moves proteins from one solution into another and is used to find the best condition to keep them stable. It’s a major bottleneck and quality concern for protein characterization, development and formulation teams. Because the process needs lots of hands-on time and eyeballing to do it right, it’s one of the most annoying things researchers have to do.

The QC duo are the ultimate tools for getting biopharmaceutical manufacturers to particle-free biologics and trouble-free drug delivery devices faster.

Particles in a drug formulation throw a wrench into development and manufacturing. Hound uses Raman spectroscopy to fingerprint protein and organic particles.  Metal, glass, and other inorganic particles get blasted by Laser-Induced Breakdown Spectroscopy (LIBS) to identify their elemental makeup. In just minutes, researchers can fully ID unwanted particles, find their source and fix their process.

Silicone oil makes a drug injection smooth and easy. Too little and the device can get jammed up. Too much and silicone can ooze into the drug and may cause aggregation. Bouncer lets manufacturers measure the thickness of sprayed-on and baked-on silicone coatings using a combo of reflectometry and interferometry. Scientists can get a handle on their coatings in minutes and fine-tune their process accordingly for top-notch device performance.

When formulating proteins at high concentrations, G22 measurements are the only way for scientists to quickly tell if a formulation makes their protein more apt to stick to itself or not. Uncle is the first platform to give researchers a G22 for their biologic with just a click of a button, letting them check for attractive forces that could cause aggregation troubles later on.

For scientists working in regulated environments, where controlling access to data and keeping its integrity are a must-have, the new 21 CFR Part 11-compliant software tools in Uncle 3.0 software can be turned on without a hitch.

High-concentration biologics don’t have to be administered as often, plus patients see better results. Lunatic’s new IgG Quant app hits the upper analysis range needed to measure these protein drugs, and does it with only 2 µL of sample straight up – no dilution needed.

Lunatic is the go-to, high-throughput tool for measuring biologic concentrations – from early research to QC. Other tools are kluge, run only a handful of samples at a time, require sample dilution which can skew results, or use way too much sample for early-stage analysis.

The new Tokyo office will be the home for sales and scientific staff, and has an applications lab ready to go for product demos.  Also, today, the company has opened its new London location, right across the street from Heathrow Airport.  This new office will be used for product development activities and product demos for customers.

Unchained Labs gets its quant on, launches Lunatic!

August 21, 2017 – Pleasanton, CA – Unchained Labs, the life sciences company that’s all about getting biologics researchers the right tool for the job, launched Lunatic today at the Bioprocessing Summit conference in Boston. Lunatic, a.k.a. The Concentration Liberator, makes batch quantification of protein, DNA and RNA a reality for biologics and genomics researchers.

Lunatic is the only UV/Vis system out there that can measure biologics in high-throughput and at high concentrations. All it takes is 2 μL and 5 minutes to measure up to 96 samples. For biologics researchers, Lunatic measures up to 150 mg/mL of their monoclonal antibodies, straight up, so they’ll never need to dilute again.

DNA and RNA samples are freaking messy. Lunatic analysis software lets genomics researchers see impurities that classic UV/Vis measurements regularly mistake as their DNA or RNA. They’ll now know if their samples are good to go or if they need another round of cleanup.

Trinean’s DropSense systems measure protein, DNA and RNA concentration in biologics and genomics samples. This is Unchained Labs’ 5th acquisition in just over 2 years.

DropSense96 is the only system out there that can measure up to 96 high concentration protein samples in 5 minutes. Scientists just load 2 microliters of sample into the proprietary DropPlate, put it in the system and that’s it. DropSense systems are a completely worry-free way to measure concentrations — no need to dilute or mix samples, clean anything before or after a run, or calibrate the system.

The proprietary, 96-well ultrafiltration consumable is designed for use on the company’s one-of-a-kind, fully automated buffer exchange systems Freeslate and Junior.

The new software turns the dial on UNcle all the way up to 11, adding 2 new applications to the 9 that were already on the first all-in-one biologics stability platform.

UNcle is now the first platform out there that has Differential Scanning Fluorimetry (DSF) and label-free thermal melting in the same box. Scientists can use the modern label-free way or choose the classic, dye-based DSF way to get their thermal melting values. With UNcle, they can now also grab the Gibbs free energy, or delta G, of biologics — the only way to quantify the stability of a protein in solution. Having these 2 new applications lets researchers do even more than they could before — like thermal stability, sizing, and aggregation measurements — with just an UNcle.

The GRUNT is the latest addition to Unchained Labs’ product line up, the 4th in less than a year.

The GRUNT unchains researchers from the time-consuming, old-school processes for making biologic formulations. Common steps like buffer prep, exchange, dialysis and sample concentration for a formulation screen can take days – and it’s all manual. With the GRUNT, scientists only spend 20 minutes on set up, picking buffers, entering parameters and loading lab wear. The GRUNT handles everything else. February 18, 2016 – Pleasanton, CA – Unchained Labs cranked its game up today, announcing its acquisition of Freeslate Inc., the leading provider of high-throughput solutions for biologics formulation development and small molecule chemistry. This is Unchained Labs’ 4th acquisition in 12 months.

Freeslate’s been partnering with Biopharma for over 20 years, developing configurable automation solutions that free scientists from major drug development bottlenecks. Their new FormPrep platform automates biologic formulation preparation and kicks out up to 12 formulations a day with only 20 minutes of prep time. Done manually, this would take 2-4 days plus a ton of hands-on time. Scientists move on to next steps way faster with FormPrep. Unchained Labs’ killer detection tools are that next step. Bringing these two biologics-focused portfolios together lets scientists analyze more biologics and formulations than ever.

July 9, 2015 – Pleasanton, CA – Unchained Labs just can’t get enough biologics stability tools, today announcing its acquisition of Avid Nano. Avid Nano designs, develops and manufactures highly sensitive dynamic light scattering (DLS) systems.

Also today, Unchained Labs UNcaged the pUNk, the world’s smallest, fastest and easiest to use protein sizing system. The pUNk is a high-performance DLS system that measures a protein’s hydrodynamic size, size distribution, aggregation population and molecular weight. Its speed and ease of use is all made possible by the low-cost BladeCell consumable, which is single use and only requires 5 µL of sample. The pUNk’s killer sensitivity comes from a proprietary optical design and distinct use of algorithms.

June 16, 2015 – Pleasanton, CA – Unchained Labs, a refreshingly new biologics tools company, UNleashed the HUNK today, the world’s first quantitative biologics stability predictor. The HUNK gives drug discovery scientists up-front insight on the stability of their protein formulations with information on how and if the safety and efficacy of their biologic will be affected during storage.

In a nutshell, the HUNK measures a time-tested thermodynamic value, ΔG, using chemical denaturation. This lets researchers determine if their protein will stay stable or aggregate over time. The kicker? The HUNK also tells them if their protein aggregates from the native or denatured state — something that can’t be done with any other tool today. Knowing which type of aggregation is the culprit gives scientists an immediate understanding of which corrective action to take. June 4, 2015 ‐ Pleasanton, CA ‐ Unchained Labs upped its game in the biologics stability market today, announcing the acquisition of AVIA Biosystems, the developer of the Isothermal Chemical Denaturation (ICD) system for measuring biologic stability. This is the second acquisition for Unchained Labs this year.

The ICD system makes something that wasn’t possible before — the routine measurement of protein stability under denaturing conditions — totally doable. It completely automates the complex sample prep and data analysis needed to make these measurements, giving formulation scientists a true walk‐away solution. The ICD system is also a perfect complement for Unchained Labs’ first product, the UNit, which simultaneously measures the top two biologic stability indicators, protein unfolding and aggregation temperatures.

May 4, 2015 – Pleasanton, CA – Unchained Labs, a refreshingly new biologic tools company, extends its coming out to this year’s PEGS conference and launches the UNit — the first and only multiplex protein stability system for biologics. PEGS: The Essential Protein Engineering Summit takes place in Boston, May 4-8, 2015.

The UNit lets drug discovery researchers look at more protein stability parameters than any other tool out there. Its 7 applications, 144 sample a day throughput and 9 µL sample size ups their knowledge base on every formulation, gets them to the right candidates exponentially faster, and uses way less sample than existing techniques. They can make better decisions, make them earlier, and skip the wasted time and effort that goes into developing candidates with no potential.

“Protein unfolding and aggregation temperatures are the first two things formulation scientists need to get a handle on. With the UNit, they can screen both at the same time, and get their hands on data from 48 samples in 2 hours,” said Tim Harkness, Founder and CEO of Unchained Labs. “There isn’t another tool that can give scientists that much actionable information on protein stability that fast.”

Pleasanton, CA February 12, 2015 – Start-up life sciences tools company Unchained Labs burst onto the scientific scene today, announcing a $25 million Series A Financing and the acquisition of Optim, the world’s first and only multiplex protein stability platform for biologics. The financing was led by Novo Ventures, Canaan Partners and TPG Biotech.

The first Unchained product, Optim, lets scientists unlock optimal biologic formulations simply and quickly. For the first time, researchers can measure multiple protein stability parameters – including denaturing temperature and aggregation onset temperature – on multiple samples simultaneously. Assays are high throughput, label-free, ultra-low volume and very easy to run.

Over 60 Optim platforms have been sold to biopharma and academic customers worldwide. Unchained Labs purchased the Optim technology from The Avacta Group. The start-up plans to continue to buy businesses and product lines and add their magic touch for developing breakthrough products and selling them like crazy.
